Summary: The compass of medicinal chemistry continues to expand with the inclusion of new technical capabilities and treatment modalities. The growth in capabilities includes the continuing improvement of screening technologies and the discovery and development of new treatment modalities. Such developments are illustrated in Volume 62 of this book series. We first highlight recent developments in the field of antibody drug conjugates (ADCs) such as noncytotoxic mechanisms of action in multiple therapeutic areas. We also illustrate the increasingly important role of biophysical screening and characterisation within medicinal chemistry workflows. Finally, the rise in popularity of the covalent mode of action is reflected in a summary of design principles, synthetic methods and screening methods for covalent fragment libraries.
